That depends a lot on the particular dealership you are ordering from. Assuming you are here in the USA, the dealer has to wait till he gets an allocation from the factory for one. Dealerships that can sell the R8 (that have certified R8 technicians) may get 1 or 2 V10 allocations per year.
So you go to a dealership and find out how long their line is. If you are lucky, there may be no line at all, or you may have to wait a year for your allocation.
Once they get an allocation, you spec the car, and it is scheduled to be built at the factory in Germany to your specifications, in the next few or several months. Then, once the car is built, you wait about 2 more months for it to be shipped and prepped and such.
Bottom line - it's not quick. Unless you don't want to spec the car and a willing to take a slightly used car for a premium - if you can find one.

Check Autotrader.com. At any given time, they list about a dozen new V10's for sale at dealers. I've called on quite a few and find about half of them are actually availalbe. (Dealers sometimes list a car even if it was ordered for a customer, they don't always remove a listing promptly after a sale, or they won't sell the car outside the local market even if they listed it.) There seem to be more R-tronics on Autotrader than manuals.
However, apparently white is this year's hot color. There have been a couple manual Ibis Whites with CF blades and black interiors that were actually for sale. Not sure about the other options.
Definitely go the Autotrader route for your car. Black, white and silver are the colors of choice for spec cars. My dealer got one allocation for a 2010, and (hooyaa!) just got another for a 2011. I will be receiving that car on approximately the one year anniversary of placing my deposit.
I need to get one that is already at a dealer as I don't have time to wait for an order.
I want either black or white with manual transmission. CF sideblade is also preferred. I need to get the car within 3 months but that should be plenty of time to find one.

If you update your profile to show where you are, others may be able to point you to some local dealers.
You can also use the Audi site to locate which dealers are R8 dealers (check the box when you do the search). Their linking and default websites for each dealer is poor. I usually use the search result for the dealer's name. Then I open a new tab and try typing in the www dealers name.com and see if I get the site or a search result for it. Once you get to the dealers real site, you can usually search their current inventory. This is another option besides the autotrader.

Autotrader has both a black and a white manual V10 listed right now. The white has silver sideblades. Don't know about the black.
I had a V8 on order in July and changed it to a V10 order in September. It is due in within a week or two. That is about 7 months from order date to delivery.
